Live attenuated varicella virus ≥1,400 PFU
Indications/Uses
Prophylaxis against varicella in individuals ≥1 yr.
Dosage/Direction for Use
SC 0.5 mL/dose. Adult & childn ≥13 yr 2 doses w/ 4-wk min interval. Childn 1-12 yr Administer 1st dose at 12-18 mth. May administer 2nd dose at 4-6 yr. Varicella outbreak Administer 2nd dose before 4 yr w/ at least 3-mth interval from 1st dose. If 1st dose is administered after 12-18 mth, 2nd dose may be administered at ≥4 yr w/ at least 3-mth interval between 1st & 2nd dose.
Contraindications
History of hypersensitivity to live attenuated varicella virus, kanamycin or erythromycin. Fever or malnutrition; CV, renal or hepatic disorder; cellular immunodeficiency; primary, acquired immunodeficiency including, immunosuppression associated w/ AIDS or clinical manifestation of HIV infection; acute myelocytic leukemia, T-cell leukemia or malignant lymphatic tumor. Highly immunosuppressed patients due to RT or intensified treatment for leukemia. History of spasm w/in 1 year prior to administration. Concomitant administration of live vaccines (oral polio, measles, rubella, mumps & BCG) w/in 1 mth. Pregnancy or willingness to conceive w/in 2 mth. Childn <12 mth.
Special Precautions
Administer epinephrine when anaphylactoid reaction occurs. Discontinue in case of administration of 6-mercaptopurine for at least 1 wk before vaccination & resume at least 1 wk after vaccination. Avoid close contact of vaccine recipients w/ immunocompromised individuals for up to 6 wk following vaccination. Immunocompromised individuals & pregnant women w/o documented history of varicella or lab evidence of prior infection. Administer w/in 72 hr after exposure to varicella in case of passive immunization by varicella-zoster Ig & done unless there are immunosuppressed symptoms. Pregnancy & lactation.
Adverse Reactions
Papule & vesicular eruptions accompanied by mild fever, herpes zoster. Inj site reactions eg, pain/soreness, swelling &/or erythema, rash, pruritus, hematoma, induration, stiffness; fever >102°F/39°C (oral), varicella-like rash (generalized or inj site).
Drug Interactions
Not to be given for at least 5 mth following blood or plasma transfusion & any Ig or varicella zoster Ig (VZIG) administration. Not to give Ig including VZIG 2 mth following vaccination. Not to be used w/ salicylates 6 wk following vaccination.
